Which is the best storyline? Let me explain how I did the above poll:
"Pilot" is the first episode ever (duh) and has the story of Sam joining Dean on the road to go find their dad and ends up hunting a Woman in White.
"Salvation" & "Devil's Trap" is a 2-part episode telling a big story of John trading the Colt to Meg while Sam & Dean go after the Yellow-Eyed Demon. Soon enough, John is captured and the boys must go save him ending the classic first season with a kick-ass 83 minute finale closing with a show-down with YED and a tragic unforeseen accident.
"In My Time of Dying" is the Season 2 premiere telling the story of Dean's out-of-body experience while in the ER. Also, Sam has to collect their weapons from the junk yard while John (unknown to Sam) is manipulating him into helping with summoning Yellow Eyes. The episode ends in an unforeseen tragic end.
"All Hell Breaks Loose, Part 1" & "All Hell Breaks Loose, Part 2" is the 2-part season finale for the show's sophomore year. It tells the tale of Sam's disappearence into a ghost town to play in a battle of wits with other telekinetic kids brought together to see who is most suitable to lead the army of the Apocalypse. The 82-minute season finale ends on a bitter/sweet note as YED is defeated, but Dean is now on the highway to hell.
"The Magnificent Seven" is the third year premiere telling a stand-alone story of the boys and Bobby hunting the Seven Deadly Sins.
"No Rest for the Wicked" is the tear-jerker finale to the third season telling the story of Dean's horrible death.
"Lazarus Rising" is the epic, unforgettable Season 4 premiere telling the tale of Dean's return and opening a can of worms by showing Sam as a changed person four months after Dean's death. Also, it introduces the series' new character named Castiel as well as Ruby's return.
"The Rapture", "When the Levee Breaks", and "Lucifer Rising" is the 3-part (124 minute long) fourth season finale tells a TON of important stories in the following order: 1.) Castiel's disappearence and return, ending with the fact that Castiel was being "schooled" in heaven on how to obey God. 2.) Dean and Bobby putting Sam into a forced rehab from demon blood by locking him in the panic room. 3.) Dean selling his soul to God. 4.) Sam's escape from the panic room. 5.) Anna goes back to heaven. 6.) Dean & Sam's split. 7.) Sam joining Ruby to killed Lilith. 8.) Castiel and Zachariah kidnapping Dean and holding him hostage. 9.) The revelation that Dean is supposed to kill Lucifer. 10.) Castiel freeing Dean. 11.) Sam killing Lilith after taking in gallons of demon blood. 12.) Lucifer being risen from the pit. 13.) The revelation that Ruby has manipulated Sam this entire time. 14.) Dean finally killing Ruby.

Aw, jeez, I can't choose.
If they were separated, then I'd say that my favourite premier would probably be IMTOD (followed closely by Lazarus Rising).
I loved IMTOD because we got to see all 3 Winchesters + Bobby, got to get a peak into each person's head, and we got a load of brotherly moments.
Favourite finale would probably be Salvation & DT. These episodes are special to me because they were my introduction to Supernatural, and I loved how much one could learn about Sam and Dean within just these 2 episodes. Dean the older brother whose desperately trying to keep his little family together, Sam the younger with so many desires and dreams of his own, but a love for his big brother that just trumps all of that. These two episodes hooked me in and this show has never let me go.
Following closely in this category are both NRFTW and Lucifer Rising. Both were brilliant, IMO.

I love them all but my faves have to Salvation/Devil's Trap for the finale. The first season was brilliant, well all the seasons are, but there was just something about the first season and I loved that all three Winchesters are working together. Plus, I totally didn't see the semi smashing the Impala coming at all.
My fave premier has to be IMOTD. IT put my emotions through the ringer. I wasn't expecting John to die and was gutted when he did. I love all that Sam did for Dean and his 'We were just starting to be brothers again' speech was so beautiful.
